Genetic and epigenetic analysis of FUT1 (fucosyltransferase 1) transgenic cloned Yorkshire piglets

Transgenic animals have been widely used in the fields of experimental and applied biology such as gene function and important proteins production studies. However, the aberrant gene expression of transgenic animals severely limits the application of transgenic technology. Copy number variation and promoter methylation changes are associated with gene expression. In the present study, we systematically analyzed the relationship between gene expression, its copy numbers, and promoter methylation level in transgenic positive, negative and natural control piglets. Our results showed that the copy numbers of the transgene have significant effect on transgene expression. We also found that the cloning piglets produced by SCNT technology can influence DNA methylation status, which can potentially effect the gene expression.
